چکیده

Cardiotrophin-1 (CT-1) is a 201 amino acid 1 member of the interleukin (IL)-6 cytokine superfamily, which includes leukemia inhibitory factor (LIF), ciliary neurotrophic factor, IL-11, and oncostatin M.2 Cardiotrophin-1 was discovered by Pennica et al2 via expression cloning of mouse embryoid bodies, using a cardiac myocyte hypertrophy screen to identify positive clones. CT-1 has hypertrophic and cardioprotective properties and acts through LIF receptor /glycoprotein 130 (gp130)-coupled signaling pathways. The LIF receptor binds CT-1, and then gp130 associates with the ligand-receptor complex and transduces the proximal signal. CT-1 intracellular signaling pathways include extracellular signal regulated kinases (ERK), mitogen activated protein (MAP) kinases, the janus kinase (JAK)/signal transducers and activators of transcription (STAT) system, and PI3-kinase/Akt. Downstream mediators of CT-1’s cellular effects include multiple ERK-coupled transcription factors, STAT-3, nuclear factorB, and heat shock proteins 56, 70, and 90. Cardiac CT-1 expression is increased by hypoxia, where it protects cardiac myocytes from ischemic and reperfusion injury and apoptosis.

Cardiotrophin-1. Review.

BACKGROUND Cardiotrophin-1 is newly discovered chemokin with a lot of functions. Aim of our work was to describe most important of them. METHODS systematically scan of available scientific resources. RESULTS Cardiotrophin-1 stimulates the proliferation of cardiomyocytes. Cardiotrophin-1 expression and plasma values are elevated in individuals with heart failure and have high diagnostic effi...

Association of cardiotrophin-1 with myocardial fibrosis in hypertensive patients with heart failure.

Cardiotrophin-1 has been shown to be profibrogenic in experimental models. The aim of this study was to analyze whether cardiotrophin-1 is associated with left ventricular end-diastolic stress and myocardial fibrosis in hypertensive patients with heart failure. Endomyocardial biopsies from patients (n=31) and necropsies from 7 control subjects were studied. Myocardial cardiotrophin-1 protein an...

Augmented expression of cardiotrophin-1 in failing human hearts is accompanied by diminished glycoprotein 130 receptor protein abundance.

BACKGROUND Cardiotrophin-1 (CT-1), a member of the interleukin-6 superfamily, is a potent inducer of cardiomyocyte hypertrophy that prolongs myocyte survival. Although cardiac CT-1 gene expression is known to be upregulated in some animal models of congestive heart failure, the activation state of the CT-1 system in patients with congestive heart failure is unknown. METHODS AND RESULTS This s...

Heart failure causes cholinergic transdifferentiation of cardiac sympathetic nerves via gp130-signaling cytokines in rodents.

Although several cytokines and neurotrophic factors induce sympathetic neurons to transdifferentiate into cholinergic neurons in vitro, the physiological and pathophysiological roles of this remain unknown. During congestive heart failure (CHF), sympathetic neural tone is upregulated, but there is a paradoxical reduction in norepinephrine synthesis and reuptake in the cardiac sympathetic nervou...

Augmented cardiac cardiotrophin-1 in experimental congestive heart failure.

BACKGROUND Cardiotrophin-1 (CT-1) is a potent hypertrophic factor discovered by coupling expression cloning in a mouse embryonic stem cell-based model of cardiogenesis. METHODS AND RESULTS The present study was designed to investigate the potential activation of atrial and ventricular CT-1 expression in pacing-induced experimental congestive heart failure (CHF) and its relationship to left ve...

Increased expression of cardiotrophin-1 during ventricular remodeling in hypertensive rats.

Cardiotrophin-1 (CT-1) stimulates longitudinal myocardial cell hypertrophy. We examined the expression of CT-1, leukemia inhibitory factor (LIF), and gp130 by competitive RT-PCR and Western blotting in Dahl salt-sensitive (DS) rats with a high-salt diet, which showed a distinct transition from left ventricular hypertrophy (LVH) to congestive heart failure (CHF). The expression levels of CT-1 mR...
